I'm using ArcMap 10.2 on a Windows 10 PC.
I tried defining the projection of the ecoregions of Alabama and Georgia shape file (USA Contiguous Albers Equal Area Conic USGS version) to my basemap (GCS North American 1983) and when I did, it didn't line up with my basemap. It moved the extent of the layer much farther then the rest of the elements and I had to full extent just to find it again.
When I first moved the shapefile into the map, it fits almost perfectly, but not exact and it's also good practice to have the same projection in each layer as well.
